Research Description

I. Characterize the SHP-l-deficient mast cells, including distribution in the lung and the function of these cells with and without stimulation. In addition, mast cells from mev mice with selective deletion of Th2 cvtokine gene will be characterized.

2. Determine whether the mev pulmonary phenotype is mast cell-dependent. Using genetic approaches we will cross breed mev mice to Kw/sh mice and examine changes in pulmonary pathology and cytokine profiles. These.studies will define whether mast cells are necessary for the allergic inflammation seen in mev mice.

3. Determine whether SHP-I deficient mast cells are able to directly induce tissue inflammation. We will use adoptive transfer technique to test this in WT and mast cell deficient mice.
